It is believed a Toyota Hilux was pulling out of a driveway at a private property when it collided with two cyclists travelling north on Mt Dandenong Tourist Road around 10.30am.
One male cyclist, a 50-year-old from Croydon, died at the scene.
The other male cyclist was taken to Royal Melbourne Hospital with minor injuries.
