Olinda-Monbulk Road is currently closed after a downed tree blocked access from both directions.
The tree fell around 10.30pm on Wednesday 1 June near Holden Road.
A Vic Roads spokesperson said it was likely the tree would be cleared by 2pm, however, with current weather it may take longer.
Local detours are in place, with Vic Roads encouraging people to take alternative routes including Monbulk Road and Perrins Creek Road, and drivers should allow for extra time.
The spokesperson said traffic is unlikely to be heavily affected.
